Rickettsia

Rickettsia are intracellular Proteobacteria associated with arthropods, including fleas, body lice, mites and ticks. The antibody response to infections by these agents is primarily directed against a lipopolysaccharide (rLPS) and 1-2 outer-membrane protein (rOmpA and/or rOmpB) antigens. The standard IFA and MIF methods detect antibody against the whole organism and remain the most commonly used of the recommended methods. Due to the range of antigens found on the whole organism, however, this broad reactivity must be compensated for by increasing the cutoff titer to 1:64-1:128. Generally, these methods discriminate between the typhus group and spotted fever group, although titers often differ between the specific infecting species and related species by 2-4-fold.

EIA techniques have ranged from whole cell lysates to individual purified antigens, with a range of effects. Our lysate-type antigens have been replaced by two current formats. The rLPS-coated plates often show the earliest antibody response and they also demonstrate the broadest reactivity within the typhus or spotted fever groups. IgM cross-reactivity to other bacteria limits this assay to IgG antibody testing.

The best data is now seen with the use of the immunodominant outer membrane proteins. With the typhus group, these antigens are rOmpB proteins from R. typhi and R. prowazekii. With the large spotted fever group the antigens are currently a mix of rOmpA and rOmpB, which show excellent group sensitivity, with added sensitivity for the particular species the antigens were isolated from.

Rickettsia MIF:

2-antigen MIF Kit (Cat No. R2-120) with Rickettsia rickettsii and Rickettsia typhi in separate microwells within each test well, for simultaneous detection of spotted fever and typhus group antibody. Antibody detection by IFA is considered group-specific, predominantly due to the presence of lipopolysaccharide (rLPS) antigens. Thus, in the 2-antigen format, both spotted fever and typhus groups are screened simultaneously in each well.
2- to 8-antigen MIF slides can be produced to your specifications. See list of strains available (below). Other antigens may be used in combination, including Coxiella burnetii, Anaplasma phagocytophilum and Francisella tularensis.

Controls are in dropper bottles containing 0.5 mL each. The Positive Control gives strong fluorescence as bottled and should titer 8-fold further. Negative Control is considered non-reactive as bottled.

Spotted Fever Rickettsia:

96-well plates (12 x 8-well strip modules) utilize purified rLPS extracted from Rickettsia rickettsii. This module is recommended for IgG screening, due to its broad reactivity with Spotted Fever Group Rickettsia.

Typhus Fever Rickettsia:

96-well plates (12 x 8-well strip modules) utilize purified rLPS extracted from Rickettsia typhi. This module is recommended for IgG screening, due to its broad reactivity with Typhus Group Rickettsia.

Rickettsia rickettsii:
